马润林，男，1958年出生，博士，研究员。功能基因组学，主要研究动物基因组的结构与功能的关系，包括构建重要动物的细菌人工染色体（BAC）文库及高密度BAC物理图谱；利用实验小鼠研究与生殖、发育相关特定基因的功能与调控，包括利用酵母双杂交技术研究蛋白相互作用及构建目标基因敲除小鼠模型；以及人类自身免疫性疾病的动物模型。

1．动物功能基因组学
有效开发一个物种的遗传资源在很大程度上取决于对其基因组结构的深入理解，并具备可以对其中任何基因进行深入研究的实验操作材料。在国家杰出青年基金的支持下，我们成功地构建了我国第一个细毛羊全基因组细菌人工染色体（BAC）文库。这是世界范围内仅有的三个绵羊BAC文库之一。 经严格检测，所构建BAC文库的库容量达到20万BAC克隆，克隆的平均DNA插入片段长度达到13万碱基（133kb），文库克隆的总碱基数达到绵羊全基因组DNA （约30亿碱基）的8.5倍。检查近400个BAC克隆，空载率小于3%。这些克隆已经PACK在近500块384孔板内供长期保存。这项工作为我国在重要经济动物全基因组高密度物理图谱这个基因组研究的关键领域做出有原创性的突破奠定了坚实基础。我们正通过杂交筛选、酶切和排列位于绵羊第20号染色体主要组织相容复合体（MHC）区段的BAC克隆，绘制该区段及临近染色体区域的BAC高密度物理图谱。
2．胚胎必须母源性因子基因MATER的功能研究
MATER是一个受精卵正常发育所必须的母源性抗原因子，我们先前的合作研究发现MATER与小鼠自身免疫性卵巢退化（AOD）有关。同时他人的研究发现缺失MATER使受精卵的发育停滞在二细胞期。我们进一步研究发现该基因在转录过程中有选择性剪切现象，形成多个mRNA转录本。这些转录本均在转录表达水平上经过RT－PCR及转录剪切体特异引物PCR扩增所证实，但是对它们在受精卵发育中的确切功能一无所知。结构预测显示MATER蛋白在结构具有Transcription Factor的特征，我们正通过Northern、Western、酵母双杂交、卵母细胞培养、免疫组化等技术研究MATER基因及选择性剪切体的确切功能与调控。
3．小鼠目标基因敲除模型
孕妇早产是造成婴儿体弱多病和死亡的主要因素之一，给家庭和社会造成种种问题和负担。但由于分娩过程在分子生物学水平的确切机制至今不明，国内外临床缺乏有效而没有副作用的调控早产药物。我们利用DNA微阵列技术研究大鼠8800多基因的表达谱时发现，约有50%基因的mRNA在大鼠妊娠的中后期至分娩前后在子宫平滑肌里表达，其中有一些是结构和功能完全未知的新基因，揭示这些新基因表达特征及功能对从分子水平上阐明分娩的确切机制有重要的意义。我们正深入调查部分所选基因的结构和组织特异时空表达特征，在此基础上设计和构建目标基因的小鼠敲除模型，以求揭示这些基因在分娩过程中的地位和作用，为寻找特异和有效的妊娠干预靶标分子以及预防早产奠定基础。
